n response to concerns expressed by a reader, we would like to offer the following comments: As indicated in the Methods of our Article, ITH14001 is a mixture of two diastereoisomers in a 1:1 ratio, as determined by NMR (see also the Supporting Information to the original Article). The remoteness of the stereocenters makes these species almost identical in physical properties. Indeed, despite many attempts, ITH14001 could not be resolved into its diastereomers using flash chromatography in a variety of hexane/ethyl acetate and dichloromethane/ethyl acetate mixtures. Crystallization in several solvents was also unsuccessfully attempted. The pharmacological evaluation of stereomixtures is well accepted, and many marketed drugs are racemic or diastereomeric mixtures.
