DOP-PCR in a chip Whole genome amplification is used to produce large quantities of DNA representative of die entire genome from limited amounts of DNA. Several strategies have been developed for this purpose, including degenerate oligonucleotide primed-polymerase chain reaction (DOP-PCR), which allows for unselected amplification of any DNA source, regardless of DNA sequence. Larry J. Kricka and co-workers at the University of Pennsylvania School of Medicine and Oak Ridge National Laboratory have taken DOP-PCR one step further by performing the reaction in a silicon-glass chip. Chip-based DOP-PCR-amplified human genomic DNA is introduced into another silicon-glass chip for subsequent assay by locus-specific multiplex PCR to detect deletions that cause Duchenne/Becker muscular dystrophy. The resulting amplicons are analyzed by traditional CE and microchip electrophoresis, and the results are compared with those using extracted high molecular weight DNA as template instead of the DOP-PCR amplified DNA Products obtained by chip-based DOPPCR were foundtobe a suitable template for multiplex PCR provided the amplicon size was
