The β-OXIDATION of a FATTY ACID begins with its ACTIVATION in a cell’s cytoplasm. ATP hydrolysis drives the addition of a COASH to form AMP and an ACYL-COA. CARNITINE transports it into the mitochondrial matrix where FAD oxidizes it to ENOYL-COA. Then a HYDRATASE adds water to make an L-β-HYDROXYACYLCOA. A DEHYDROGENASE uses NAD+ to oxidize this to a β-KETOACYL-COA. Finally, THIOLASE cleaves it, making a shorter acyl-CoA that recycles, and an ACETYLCOA. This drives the KREBS CYCLE, and the FADH2 and NADH produced here and above drive ELECTRON transport making lots of ATP, which is the point, after all. Abnormal states like DIABETES and STARVATION lead to ketosis, in which the KETONE BODIES, ACETOACETATE, 3HYDROXYBUTYRATE, and ACETONE, are made. FATTY ACIDS ENERGIZE US.
