A variety of derivatives of 2-amino-5-nitrothiazole and 2-hydrazino-5-nitrothiazole were prepared as potential schistosomicides. The general lack of activity over a wide range of structural types serves to emphasize the specificity surrounding the highly active l-(5-nitro-2-thiazolyl)-2-imidazolidinone(11).

A variety of nitro heterocyclic systems have been demonstrated to possess useful therapeutic properties. n'itrofurans for example have been extensively investigated' and several are useful clinically in man. More recently nitroimidaeoles2 have found a place in the treatment of protozoan infections. Derivatives of nitrothiazole have remained relatively unexplored. I t has been known for some time that simple analogs of 2amino-5-nitrothiazole are effective against trichomonad infections. I n 1955 the activity of acetamide (I) against

dride gave a product which appears to be VI1 or the isomeric TrIII. This material which exhibits a strong ester carbonyl at 1730 cm-I is apparently formed from the imide during the purification from ethanol. Phthalic anhydride provided only the imide I X in poor yield.

experimental infections of Scliistosoma inansoni in mice was d e m ~ n s t r a t e d . ~Several years ago Lambert, Wilhelm, and coworkers4-' reported their success in the modification of this system culminating in the introduction of a new clinical agent for schistosomiasis, niridasole (11). We report here some investigations into the chemistry of 2-amino-5-nitrothiaaole which have been proceeding in our laboratories during the past several years, and the activities of the products of this work as antiparasitic agents. Treatment of 2-amino-5-nitrothiazole (AXT) with simple dicarboxylic acid anhydrides led to bisamides or half-amide-half-acids. Thus succinic anhydride, glut aric anhydride, 1,2-cyclohexanedicarboxylic anhydride, and cis-4-cyclohexene-l,2-dicarboxylicanhydride provided 111-VI. 1,2,4,5-Benzenetetracarbox ylic anhy-

Treatment of AXT with oxalyl chloride in pyridine gave the bisamide X. The use of longer chain dicarboxylic acid chlorides led directly to the desired imides.
