To c o n t r o l t h e heavy l o s s e s o f c r o p s from i n s e c t s and o t h e r p e s t s , t h e use o f d i f f e r e n t p e s t i c i d e s has become e x t e n s i v e , and w i t h t h e p a s s i n g o f t i m e , more o f t h e s e compounds a r e b e i n g d e v e l o p e d and p r o d u c e d on a c o m m e r c i a l s c a l e . At p r e s e n t , a p a r t from t h e i n o r g a n i c compounds s u c h as z i n c p h o s p h i d e and a r s e n i o u s o x i d e , s e v e r a l h u n d r e d o r g a n i c compounds, h a v i n g w i d e l y d i f f e r e n t c h e m i c a l c o n s t i t u e n t s and p r o p e r t i e s , a r e b e i n g u s e d t h r o u g h o u t the w o r l d . The f r e q u e n t use o f t h e s e o r g a n i c p e s t i c i d e s has r e s u l t e d i n t h e i r widespread d i s t r i b u t i o n i n the environment. Often they are found i n unwanted p l a c e s , t h e r e b y c a u s i n g p o l l u t i o n p r o b l e m s . B e c a u s e t h e s e p e s t i c i d e s a r e t o x i c n o t o n l y t o i n s e c t s and p e s t s b u t a l s o , i n v a r y i n g d e g r e e s , t o h i g h e r a n i m a l s and m a n , t h e s e c h e m i c a l s have c r e a t e d a p u b l i c h e a l t h m e n a c e . Today many c o m m e r c i a l p r o d u c t s a r e a v a i l a b l e as home remedi e s a g a i n s t f l i e s , t i c k s , b u g s , r a t s and o t h e r i n s e c t s and p e s t s under t h e t r a d e names o f ' F l i t ' , T i c k - 2 0 , B u g m a r , D a l f , B u g t o n and many m o r e . These p e s t i c i d e s a r e a l s o a v a i l a b l e i n c o n c e n t r a ted s o l u t i o n s , powders, a e r o s o l s , e m u l s i o n s , e t c . f o r a g r i c u l tural purposes. Due t o t h e i r e a s y a v a i l a b i l i t y and r a p i d t o x i c a c t i o n , t h e s e p e s t i c i d e s have been m i s u s e d as h o m i c i d a l a g e n t s and a c t e d as a c c i d e n t a l p o i s o n s . The m i s u s e has become a p r o b l e m f o r t h e t o x i c o l o g i s t , e s p e c i a l l y i n I n d i a , where p o i s o n i n g s by t h e u s e o f p e s t i c i d e s a r e i n c r e a s i n g a n n u a l l y . The i n v e s t i g a t i o n o f t h e s e a c c i d e n t a l d e a t h s has become a g r e a t c h a l l e n g e t o t h e forensic toxicologist. In o r d e r t o e s t a b l i s h t h a t t h e d e a t h o f a v i c t i m i s due t o t h e i n g e s t i o n o f a s p e c i f i c p o i s o n , t h e a b s o l u t e p r o o f needed i s t h e d e t e c t i o n and i d e n t i f i c a t i o n o f t h e p o i s o n i n v a r i o u s o r g a n s of the body. This a n a l y s i s p r o v i d e s t h e d a t a on t h e d i s t r i b u t i o n o f t h e p o i s o n i n d i f f e r e n t o r g a n s and may be o f g r e a t v a l u e i n d e d u c i n g t h e p r o b a b l e o r i g i n a l d o s e i n g e s t e d a n d a l s o t h e t i m e , a n d t o some e x t e n t , the nature o f a d m i n i s t r a t i o n o f the poison. These d a t a may a l s o h e l p t o g i v e a c l u e r e g a r d i n g t h e cause o f d e a t h . However, f r o m e x p e r i e n c e i t has been f o u n d t h a t t h e knowledge o f ( 1 ) t h e a c t i o n o f t h e p e s t i c i d a l p o i s o n under c o n s i d e r a t i o n , ( 2 ) c l i n i c a l c o u r s e o f t h e p o i s o n i n g a n d ( 3 ) t h e postmortem f i n d i n g s i s o f great importance i n the i n t e r p r e t a t i o n o f the r e s u l t s obtained from chemical a n a l y s i s o f t h e m a t e r i a l r e c e i v e d . T h e r e a r e o c c a s i o n s i n w h i c h d e a t h i s u n d o u b t e d l y due t o a p e s t i c i d e , and y e t t h e c h e m i c a l a n a l y s i s f a i l s t o r e v e a l i t s p r e s e n c e o r shows o n l y i n s i g n i f i c a n t t r a c e s . T h i s may be due t o the r a p i d m e t a b o l i c conversion o f the p a r t i c u l a r p e s t i c i d e . This i s the case w i t h p a r a t h i o n which i s r a p i d l y converted t o p a r a n i t r o p h e n o l due t o i n v i v o h y d r o l y s i s . Pesticides, especially t h o s e t h a t a r e c o m p a r a t i v e l y l e s s t o x i c t o mammals, s u c h a s d i c h l o r o v o s , e t c , may be e x c r e t e d r a p i d l y a n d , t h e r e f o r e , may p e r h a p s be f o u n d i n t h e u r i n e , t h o u g h n o t i n t h e t i s s u e s . The s t u d y o f t h e d i s t r i b u t i o n o f a p o i s o n , p o s t m o r t e m , p r e s e n t s a complex problem. The e s t i m a t i o n o f t h e p o i s o n i n t h e b l o o d p r e s e n t s a c o m p l e x p r o b l e m due t o t h e p u t r e f a c t i v e changes i n b l o o d and t h e minute q u a n t i t y a v a i l a b l e f o r a n a l y s i s . Distrib u t i o n o f i n s e c t i c i d a l p o i s o n s from t h e b l o o d t o t h e t i s s u e s and body f l u i d s t a k e s p l a c e w i t h some r e c i r c u l a t i o n t o t h e b l o o d ( l _ ) . The body t i s s u e s a r e g e n e r a l l y s u b m i t t e d t o a t o x i c o l o g i c a l l a b o r a t o r y i n s u f f i c i e n t q u a n t i t i e s ; hence t h e s e s p e c i m e n s a r e better suited for quantitative analysis. The d e t e c t i o n a n d d e t e r m i n a t i o n o f t h e unchanged i n s e c t i c i d e i n t h e s t o m a c h , i n t e s t i n e , l i v e r , k i d n e y , s p l e e n , l u n g s , b r a i n , and i n o t h e r t i s s u e s and body f l u i d s a r e u s e f u l i n d i c a t o r s o f d i r e c t i n s e c t i c i d e action. I n o r d e r t o r e a c h a c o n c l u s i o n when and how a n d , a n d t o some e x t e n t , how much o f a p a r t i c u l a r i n s e c t i c i d e was a d m i n i s t e r e d , t h e f o r m and q u a n t i t y i n w h i c h t h e i n s e c t i c i d e e x i s t s i n d i f f e r e n t t i s s u e s a n d i n b l o o d a n d u r i n e may g i v e v a l u a b l e c l u e s . I n d e e d , a n i n t e l l i g e n t d e d u c t i o n i s a l s o b a s e d on t h e knowledge o f m e t a b o l i c pathways a n d f o r m a t i o n o f o t h e r d e r i v a t i v e s . Thus, as p a r a t h i o n i s e x c r e t e d i n t h e u r i n e u l t i m a t e l y as p - n i t r o p h e n o l , u r i n a r y p - n i t r o p h e n o l l e v e l s may i n d i c a t e t h e e x t e n t o f exposure t o p a r a t h i o n ( 2 j . In c a s e s o f m i l d a n d m o d e r a t e e x p o s u r e , t h e e x c r e t e d p - n i t r o p h e n o l i n u r i n e was f o u n d t o be o f t h e o r d e r o f 0 . 0 5 7 t o 0 . 3 2 2 mg. p e r c e n t . In s e v e r e a n d f a t a l c a s e s o f p o i s o n i n g by p a r a t h i o n , t h e l e v e l o f p - n i t r o p h e n o l i n u r i n e was f r o m 0 . 1 6 t o 1.16 mg. p e r c e n t . para-Nitrophenol thus i s r a p i d l y e x c r e t e d i n u r i n e a n d no l o n g e r d e t e c t e d 4 8 h o u r s a f t e r the exposure t o the p e s t i c i d e .

The methods o f e x t r a c t i o n o f t h e p e s t i c i d e s f r o m t i s s u e s a s r e p o r t e d by t h e a u t h o r and coworkers (13-15) a r e d e s c r i b e d b e l o w : I s o l a t i o n o f O r g a n o p h o s p h o r u s I n s e c t i c i d e s From Body T i s s u e s . Twenty grams o f t h e t i s s u e i s m a c e r a t e d i n t o a f i n e s l u r r y b y m i x i n g i t w i t h e q u a l amounts o f a n h y d r o u s s o d i u m s u l f a t e a n d p l a c i n g i t i n t o a c o n i c a l f l a s k f i t t e d w i t h an a i r c o n d e n s e r . F i f t y m i l l i l i t e r s o f n-hexane i s a d d e d , and t h e f l a s k i s heated on a b o i l i n g w a t e r b a t h f o r o n e h o u r . A f t e r c o o l i n g , t h e n hexane l a y e r i s f i l t e r e d . The e x t r a c t i o n o f t h e r e s i d u a l s l u r r y i s repeated twice with twenty-five m i l l i l i t e r portions o f n hexane. The f i l t e r e d n - h e x a n e f r a c t i o n s a r e c o m b i n e d a n d t r a n s fered to a separatory funnel. The hexane l a y e r i s v i g o r o u s l y s h a k e n f o r f i v e m i n u t e s s u c c e s s i v e l y w i t h 15 m l - , 10 m l - , a n d 10 m l - p o r t i o n s o f a c e t o n i t r i l e p r e v i o u s l y s a t u r a t e d w i t h n-hexane. The a c e t o n i t r i l e l a y e r s a r e m i x e d a n d p o u r e d i n t o a n o t h e r s e p a r a tory funnel. I n c a s e s o f d i m e t h o a t e , D a s a n i t , phosphamidon a n d DDVP p o i s o n i n g s , t h e t h r e e a c e t o n i t r i l e f r a c t i o n s a r e combined and e v a p o r a t e d t o d r y n e s s , a n d t h e r e s i d u e i s t a k e n up i n one m i l l i l i t e r o f acetone. Further extraction of acetonitrile i s a v o i d e d a s t h i s r e s u l t s i n a p p r e c i a b l e l o s s o f p e s t i c i d e s . The a c e t o n i t r i l e l a y e r i s d i l u t e d w i t h w a t e r t o t e n times o f i t s o r i g i n a l volume. T w e n t y - f i v e m l . o f s a t u r a t e d sodium s u l f a t e s o l u t i o n i s added t o t h i s s o l u t i o n a n d e x t r a c t e d t h r e e t i m e s w i t h t w e n t y - f i v e m i l l i l i t e r - p o r t i o n s o f n-hexane p r e v i o u s l y s a t u r a t e d with d i s t i l l e d water. T h i r t y grams o f a n h y d r o u s sodium s u l f a t e and f i f t y m i l l i l i t e r s o f acetone a r e added. The c o n i c a l f l a s k i s p l a c e d on a b o i l i n g w a t e r b a t h a t 80° f o r h a l f an h o u r . The f l a s k i s removed f r o m t h e w a t e r b a t h , c o o l e d t o room t e m p e r a t u r e , and t h e c o n t e n t s a r e f i l t e r e d . The p r o c e s s i s repeated twice with twenty-five m i H i l i t e r - p o r t i o n s of acetone. The f i l t e r e d a c e t o n e l a y e r s a r e combined and t r a n s f e r e d t o a séparatory f u n n e l . One h u n d r e d m i l l i l i t e r s o f d i s t i l l e d w a t e r and t w e n t y m i l l i l i t e r s o f a s a t u r a t e d aqueous s o l u t i o n o f s o d i u m s u l f a t e i s added t o t h e a c e t o n e l a y e r . The s o l u t i o n i s t h e n extracted with chloroform previously saturated with d i s t i l l e d w a t e r u s i n g t w e n t y and two t e n m i l l i l i t e r - p o r t i o n s . The e x t r a c t i o n i s c o m p l e t e d by s l o w l y s h a k i n g f o r f i v e m i n u t e s (20 t o 25 shakings per m i n u t e ) . The s e p a r a t e d c h l o r o f o r m l a y e r s a r e c o m b i n e d and p l a c e d i n a n o t h e r 250-ml s e p a r a t o r y f u n n e l . Twenty m i l l i l i t e r s o f 5% aqueous p o t a s s i u m h y d r o x i d e s o l u t i o n i s added and s h a k e n f o r one m i n u t e . The c h l o r o f o r m l a y e r i s s e p a r a t e d by d i s c a r d i n g t h e aqueous l a y e r a n d a g a i n s h a k e n f o r one m i n u t e w i t h t w e n t y mi H i l i t e r s o f 5% KÛH. The s e p a r a t e d c h l o r o f o r m l a y e r i s now washed w i t h f i v e 20 m l . p o r t i o n s o f d i s t i l l e d w a t e r by s l o w l y s h a k i n g i t f o r one m i n u t e a t each t u r n . The washed c h l o r o f o r m l a y e r i s d r i e d by p a s s i n g t h r o u g h a l a y e r o f a n h y d r o u s s o d i u m s u l f a t e p r e v i o u s l y wetted with dry chloroform. The o r g a n i c s o l v e n t i s t h e n c o n c e n t r a t e d up t o two m i l l i l i t e r s on a warm w a t e r b a t h a t 6 0 ° , and t h e r e m a i n i n g s o l v e n t i s e v a p o r a t e d t o d r y n e s s i n a c u r r e n t of dry a i r . The r e s i d u e w h i c h c o n t a i n s t h e p e s t i c i d e i s t h e n d i s s o l v e d i n one m i l l i l i t e r o f a c e t o n e . Methods o f A n a l y s i s o f P e s t i c i d e s . The e x t r a c t e d r e s i d u e o b t a i n e d a f t e r i s o l a t i o n f r o m t i s s u e s and o t h e r b i o l o g i c a l m a t e r i a l s i s s u b j e c t e d t o q u a l i t a t i v e and q u a n t i t a t i v e d e t e r m i n a t i o n o f the p e s t i c i d e s . S o m e t i m e s , t h e amount o f m a t e r i a l a v a i l a b l e i s so s m a l l t h a t t h e c o l o r i m e t r i c and o t h e r a l l i e d methods c a n n o t be s u c c e s s f u l l y a p p l i e d as some o f t h e r e s i d u e i s l i k e l y t o be l o s t d u r i n g t h e p u r i f i c a t i o n t e c h n i q u e . Furthermore, these p u r i f i c a t i o n techniques required for spectrophotometry, c o l o r i m e t r y , and o t h e r s o p h i s t i c a t e d i n s t r u m e n t a l methods a r e a p p r e c i a b l y t i m e c o n s u m i n g . T h e r e f o r e , o t h e r t e c h n i q u e s were s o u g h t f o r the q u a n t i t a t i v e d e t e r m i n a t i o n of p e s t i c i d e s . T h i n l a y e r chromat o g r a p h i c (TLC) t e c h n i q u e s were f o u n d t o be most s u i t a b l e f o r toxicological analysis of pesticides. Randerath(lJ5) s t a t e d t h a t

a c c u r a t e r e s u l t s i n q u a n t i t a t i v e TLC were o b t a i n e d by r e m o v a l o f t h e s u b s t a n c e t o be a n a l y z e d f r o m t h e a d s o r b e n t u s i n g s u i t a b l e solvents f o r elution. C h o u 1 i s ( 1 7 , 1 8 ) has a l s o advocated t h e e l u t i o n technique f o r q u a n t i t a t i v e analyses. The e l u t i o n t e c h n i q u e i s n o r m a l l y used i n a s s o c i a t i o n w i t h g r a v i m e t r i c , s p e c t r o p h o t o m e t r y , c o l o r i m e t r i c , f l u o r o m e t r i c and other e s t i m a t i o n methods(19). The g r a v i m e t r i c method b e i n g u n r e l i a b l e and y i e l d i n g g e n e r a l l y h i g h r e s u l t s i s n o t s u i t a b l e for toxicological analysis. The a c c u r a c y o f t h e f l u o r o m e t r i c method i s g e n e r a l l y c o n s i d e r e d t o be l o w . R e g a r d l e s s , t h e f l u o r o m e t r i c method c a n o n l y be a p p l i e d t o t h o s e s u b s t a n c e s w h i c h f l u o r e s c e under t h e assay c o n d i t i o n s o r which f l u o r e s c e a f t e r r e a c t i n g w i t h some s u i t a b l e r e a g e n t . C o l o r i m e t r i c methods a r e a p p l i c a b l e o n l y t o those s u b s t a n c e s which form c o l o r e d s o l u t i o n s w i t h a c h r o m o g e n i c r e a g e n t , t h e a b s o r b a n c e o f w h i c h c a n be measured using a spectrophotometer o r c o l o r i m e t e r . These e l u t i o n methods p r e s e n t s e v e r a l d i s a d v a n t a g e s f o r t h e f o l l o w i n g r e a s o n s : ( i ) I n a c c u r a t e m a r k i n g o f t h e s p o t t o be e l u t e d f r o m TLC p l a t e , ( i i ) Incomplete d e s o r p t i o n o f t h e s u b s t a n c e i n t h e s o l v e n t used for elution. ( i i i ) I n t e r f e r e n c e due t o c e r t a i n s u b s t a n c e s e x t r a c t e d f r o m t h e adsorbents. For these r e a s o n s , Purdy and Truter(2fJ) and s e v e r a l o t h e r w o r k e r s have s t r e s s e d methods by w h i c h measurement o f t h e a r e a o f t h e d e v e l o p e d c o l o r e d s p o t on TLC p l a t e s c a n be u s e d d i r e c t l y f o r quantitative analysis. Methods f o r m e a s u r i n g t h e s p o t a r e a may be c l a s s i f i e d as f o l l o w s : 1. V i s u a l comparison o f spot i n t e n s i t y and/or s i z e t o a s t a n d a r d amount o f t h e same p e s t i c i d e r u n on t h e same TLC p l a t e . This p r o v i d e s a n e s t i m a t i o n o f t h e s u b s t a n c e w i t h a p p r o x i m a t e l y 20% a c c u r a c y . A d e t a i l e d d e s c r i p t i o n o f t h i s method o f v i s u a l e s t i m a t i o n o f t h i n l a y e r c h r o m a t o g r a p h s has been g i v e n by J o h n s o n ( 2 1 ) , who h a s v e r y w i s e l y commented t h a t i t i s b e t t e r t o be " a p p r o x i mately r i g h t than p r e c i s e l y wrong". 2. The a r e a o f t h e s p o t may be t r a c e d o n g r a p h p a p e r , a n d t h e s q u a r e s on t h e p a p e r c o u n t e d a n d compared w i t h t h o s e o f known amounts o f a s t a n d a r d p e s t i c i d e . T h i s method g i v e s s l i g h t l y b e t t e r r e s u l t s than 1., but l a c k s accuracy. 3. S c a n n i n g t h e d e v e l o p e d c o l o r e d s p o t on t h e t h i n l a y e r chromatographic plate with the help o f a p h o t o e l e c t r i c d e n s i tometer. G a e n s h i r t and Polderman(22) have mentioned t h a t t h e e r r o r s o f t h e q u a n t i t a t i v e d e t e r m i n a t i o n o f a p e s t i c i d e by p l a n i m e t r i c measurements o f t h e a r e a o f t h e s p o t s a r e g e n e r a l l y h i g h a s a r e s u l t o f v a r i a t i o n s i n t h e shape o f t h e s p o t a n d d i f f i c u l t i e s i n defining the outline o f the spot. M o r r i s o n and C h a t t e n ( 2 3 ) and O s w a l d a n d F l u e c k ( 2 4 ) have p o i n t e d o u t t h e same d i f f i c u l t i e s i n t h e v i s u a l measurement o f t h e a r e a o f t h e s p o t . Quantitative a n a l y s i s on TLC w i t h a f l y i n g s p o t d e n s i t o m e t e r has been s u g g e s t ed by S c h u t e a n d c o - w o r k e r s ( 2 5 ) . Genest(27) used a P h o t o v o l t d e n s i t o m e t e r f o r d e t e r m i n a t i o n o f d r u g s on T L C . Later, B l u n d e n and c o - w o r k e r s ( 2 8 ) , S h e l l a r d ( 2 9 ) and Thomas e t a h (30) a l s o r e p o r t e d on t h e use o f a d e n s i t o m e t e r f o r q u a n t i t a t i o n . S h e l l a r d and A l a m ( 3 1 J i n 1968 have made a d e t a i l e d s t u d y o f t h e d e n s i t o m e t r i c method f o r TLC. S e v e r a l w o r k e r s have d e s c r i b e d t h e t h i n l a y e r c h r o m a t o g r a p h i c technique f o r the d e t e r m i n a t i o n o f organophosphorus p e s t i c i d e s ( 3 2 - 4 4 ) a s w e l l as c h l o r i n a t e d p e s t i c i d e s ( 4 5 - 5 6 ) . V y l e g z h a n i n a and Kalrrrykova(57) have d e s c r i b e d t h e use o f TLC f o r d e t e r m i n a t i o n o f DDVP i n p l a n t s , s o i l and w a t e r . Determin a t i o n o f o r g a n o p h o s p h o r u s i n s e c t i c i d e s i n r i v e r w a t e r and e f f l u e n t s have a l s o been used by A s k e w , e t a h ( 5 8 ) . Z y c i n s k i ( 5 9 j has d e t e r m i n e d t h e c o n c e n t r a t i o n o f m a l a t h i o n , DDVP a n d o t h e r o r g a n o p h o s p h o r u s i n s e c t i c i d e s i n w a t e r by d i r e c t measurement on TLC plates. TLC d e t e r m i n a t i o n o f s e v e r a l o r g a n o p h o s p h o r u s i n s e c t i c i d e s s u c h as f e n i t r o t h i o n , f e n t h i o n , and m a l a t h i o n i n s o i l has been s u g g e s t e d by Cywi η s ka-Smo t e r ( 6 0 ) u s i n g Congo r e d s p r a y r e a g e n t f o r t h e d e v e l o p m e n t o f t h e s p o t s on T L C . Several workers ( 6 1 , 6 2 , 6 3 , 6 4 ) have u t i l i z e d t h e e s t e r a s e i n h i b i t i o n t e c h n i q u e f o r t h e l o c a t i o n o f t h e o r g a n o p h o s p h o r u s i n s e c t i c i d e s on TLC and t h e i r d i r e c t measurement on t h e p l a t e s . Routine determination of o r g a n o p h o s p h o r u s i n s e c t i c i d e s by TLC has a l s o been s u g g e s t e d by A n t o i n e and M e e s ( 3 9 ) . They have s u g g e s t e d p a l l a d i u m c h l o r i d e s p r a y o r 4 - ( 4 - n i t r o b e n z y l ) p y r i d i n e f o r l o c a t i o n o f t h e s p o t s on t h e TLC p l a t e and c o m p a r i s o n o f i n t e n s i t y and a r e a o f t h e d e v e l oped c o l o r e d s p o t s w i t h t h o s e o f known amounts o f t h e i n s e c t i c i d e s on t h e same TLC p l a t e . He11omann(65) has u s e d b o v i n e l i v e r e x t r a c t as s p r a y f o l l o w e d by c o m p a r i s o n o f t h e d e v e l o p e d y e l l o w c o l o r e d soot f o r the s e m i - q u a n t i t a t i v e d e t e r m i n a t i o n . Kosmatyi e t aj_. (66) have u s e d b r o m o p h e n o l - b l u e / s i l v e r n i t r a t e as c h r o m o g e n i c r e a a e n t f o r t h e d e t e r m i n a t i o n o f d i a z i n o n . The a u t h o r ( 6 7 , 6 8 , 6 9 ) has r e p o r t e d t h e d e n s i t o m e t r i c d e t e r m i n a t i o n o f m i c r o gram amounts o f o r g a n o p h o s p h o r u s i n s e c t i c i d e s i n t i s s u e s . D e t e r m i n a t i o n o f d i m e t h o a t e and m a l a t h i o n u s i n g p a l l a d i u m c h l o r i d e s p r a y on TLC p l a t e was d e s c r i b e d by M a c N e i l a n d c o workers (70). Q u a n t i t a t i v e d e t e r m i n a t i o n of organophosphorus p e s t i c i d e s by TLC d e n s i t o m e t r y 71J a n d TLC f l u o r o m e t r y ( 7 2 ) have a l s o been d e s c r i b e d . I v e y and 0 e h l e r ( 7 3 ) have d e s c r i b e d t h e d e t e r m i n a t i o n o f some o r g a n o p h o s p h o r u s p e s t i c i d e s i n t i s s u e s a n d urine of c a t t l e . G e t z ( 7 4 ) has d e s c r i b e d t h e q u a n t i t a t i v e e s t i m a t i o n o f c h l o r i n a t e d p e s t i c i d e s by d e n s i t o m e t r y . Dobbins(75^) has used t h e d e n s i t o m e t r i c method f o r q u a n t i t a t i o n o f c h l o r i n a t e d p e s t i c i d e s using s i l v e r n i t r a t e spray reagent. Q u a n t i t a t i v e TLC t e c h n i q u e f o r o r g a n o c h l o r o p e s t i c i d e s was a l s o s u g g e s t e d by M a c N e i l e t a h ( 5 1 J and L a k s h i n a r a y a n a and c o - w o r k e r s ( 7 6 ) . Quan t i t a t i o n o f c a r b a m a t e p e s t i c i d e s by f l u o r e s c e n c e d e n s i t o m e t r y on TLC was d e s c r i b e d by some o f t h e w o r k e r s ( 7 7 , 7 8 , 7 9 , 8 0 ) . G l a s s p l a t e s ( 2 0 x 2 0 c m . ) a r e w e l l c l e a n e d w i t h non g r e a s y s o l v e n t a n d a r r a n g e d on t h e a l i g n i n g t r a y . T w e n t y - f i v e grams o f the adsorbent s i l i c a gel-G (or alumina with calcium s u l f a t e b i n d e r ) a r e mixed w i t h f i f t y m i l l i l i t e r s o f d i s t i l l e d water i n a 250 m l - f l a s k a n d g e n t l y s h a k e n t o f o r m a homogeneous s l u r r y . T h i s s l u r r y i s t r a n s f e r r e d i m m e d i a t e l y ( w i t h i n s i x t y seconds o f mixing adsorbent w i t h water) t o the a p p l i c a t o r . The s p r e a d e r i s a d j u s t e d f o r a 250-um t h i c k l a y e r ( o r o t h e r s u i t a b l e t h i c k n e s s ) b e f o r e h a n d , and a t h i n l a y e r o f t h e a d s o r b e n t i s c o a t e d on t h e plates. I n t h i s way a homogeneous 250-ym t h i c k l a y e r o f t h e absorbent i s applied to the glass p l a t e s . The p l a t e s a r e d r i e d a t room t e m p e r a t u r e a n d t h e n a c t i v a t e d i n a n oven a t 110° f o r 30 minutes. Storage. The p l a t e s a r e s t o r e d i n a d e s i c c a t o r o v e r a n h y drous c a l c i u m s u l f a t e and p r o t e c t e d from t h e l a b o r a t o r y fumes. A p p l i c a t i o n o f samples. An a l i q u o t ( a b o u t one t o t e n y l ) o f s o l u t i o n o f t h e sample i s s p o t t e d on t h e p l a t e w i t h a c a p i l l a r y d r o p p e r o r m i c r o p i p e t t e 1.5 cm. f r o m one e n d o f t h e p l a t e . Care must be t a k e n t o a v o i d d i f f u s i o n o f t h e s p o t ; m o r e o v e r , t h e a r e a o f t h e s p o t s h o u l d be r e s t r i c t e d t o a t w o - o r t h r e e m i l l i m e t e r diameter. To e n s u r e t h i s , t h e s o l u t i o n s h o u l d be a p p l i e d i n s t a g e s a n d d r i e d i n a s t r e a m o f warm a i r . The l a y e r on t h e p l a t e s h o u l d n o t be p e n e t r a t e d d u r i n g s p o t t i n g . M u l t i p l e spots should be a p p l i e d on t h e p l a t e a t l e a s t one c e n t i m e t e r a p a r t . Development o f t h e p l a t e . A f t e r ensuring that the applied spots are d r y , the spotted plate i s placed i n a developing chamber c o n t a i n i n g t h e s o l v e n t s y s t e m and t h e chamber i s c l o s e d w i t h a n a i r - t i g h t l i d . The s o l v e n t s h o u l d be a b o u t 0 . 5 t o 1 c m . deep i n t h e d e v e l o p i n g c h a m b e r . Some o f t h e compounds a r e d e t e c t e d on t h e p l a t e by v i e w i n g t h e p l a t e u n d e r u l t r a v i o l e t l i g h t (254 nm). Organochloride. Densitometry
Densitometry i s a technique o f d i r e c t d e t e r m i n a t i o n o f the c o n c e n t r a t i o n o f t h e c o l o r e d d e r i v a t i v e on t h e c h r o m a t o g r a m by absorptometry. Discussion From t h e p e r u s a l o f d a t a r e c o r d e d i n T a b l e s I I I a n d I V , i t i s evident that i n s e c t i c i d e s are d i s t r i b u t e d i n p r a c t i c a l l y a l l the v i s c e r a l t i s s u e s . H o w e v e r , t h e amount o f i n s e c t i c i d e i n d i f f e r e n t body t i s s u e s depends o n t h e t i m e p e r i o d between i n g e s t i o n o f t h e i n s e c t i c i d e and d e a t h . A t t h e i n i t i a l o n s e t o f symptoms, t h e m a j o r i t y o f t h e i n s e c t i c i d e s i s s t i l l i n t h e stomach. But i n c a s e s where d e a t h i s p r o l o n g e d , t h e i n s e c t i c i d e a c c u m u l a t e s i n o t h e r t i s s e s , e . g . l i v e r and k i d n e y . By c o n d u c t i n g c o n t r o l e x p e r i m e n t s on w h i t e r a t s w h i c h were f e d l e t h a l d o s e s o f some o f t h e o r g a n o p h o s p h o r u s i n s e c t i c i d e s ( p a r a t h i o n and m a l a t h i o n ) i t was o b s e r v e d t h a t t h e s e i n s e c t i c i d e s t e n d e d t o accumul a t e i n t h e body f a t , a n d a s m a l l amount was e x c r e t e d i n t h e urine. A s m a l l amount was a l s o e l i m i n a t e d t h r o u g h e x c r e t a . A h i g h e r c o n c e n t r a t i o n o f t h e i n s e c t i c i d e i n t h e stomach i s i n d i c a t i v e t h a t t h e death o f t h e v i c t i m took place w i t h i n a s h o r t p e r i o d , b e c a u s e t h e i n s e c t i c i d e c o u l d n o t even be c o m p l e t e l y e l i m i n a t e d from t h e stomach. A c o m p a r a t i v e l y l a r g e c o n c e n t r a t i o n o f p e s t i c i d e i n l i v e r t i s s u e i s s u g g e s t i v e o f a d e l a y i n death from t h e t i m e o f administration of the insecticide. Abstract The absolute proof of fatal cases of poisoning is the detection of the poison in different organs of the body. Due to the chemical nature of the poison and its metabolism, the suspected chemical is rarely evenly distributed in body tissues but tends to accumulate in some particular organ. Hence, to conduct a thorough forensic investigation, it is necessary to analyze different visceral tissues qualitatively and quantitatively for poison residues. In this paper, the quantitative TLC technique using densitometry for the determination of traces of pesticides has been described in detail. The concentration of pesticides in various human organs from poisoning cases has been recorded as well as data on tissue distribution. The distribution pattern of several important pesticides, like parathion, malathion, sumithion, and endrin has been studied. The results of these studies have aided in solving the questions in the probable causes of death or poisoning-- how and when the pesticide was probably administered, and i f death occurred by suicide, homicide or accident. Several actual cases of poisoning and subsequent tissue analyses by quantitative TLC have been described.
