The mechanisms for the brassinosteroid antistress effect i n plants were investigated. In wheat leaves under heat shock (40ºC) 22S,23S-homobrassinolide and 24-epibrassinolide activated t o t a l protein synthesis and induced de novo polypeptide synthesis at normal and high temperatures. Molecular weight determinations showed that some of the proteins induced by brassinosteroids at normal temperature corresponded to heat shock proteins. 22S,23S-homobrassinolide also stimulated formation of heat shock granules i n the cytoplasm and increased thermotolerance of t o t a l protein synthesis under heat shock. 24-epibrassinolide produced a protective effect on leaf c e l l ultrastructure i n leaves placed under saline stress (0.5 M NaCl) and prevented n u c l e i and chloroplast degradation. Brassinosteroids ( B S s ) r e p r e s e n t a new g r o u p o f p l a n t hormones t h a t p o s s e s s a b r o a d s p e c t r u m o f p h y s i o l o g i c a l a c t i v i t i e s ( 1 , 2 ) . A most i n t r i g u i n g p r o p e r t y o f B S s i s t h e i r capacity t o increase stress resistance i n plants, b u t t h e mechanism o f s u c h a n a n t i s t r e s s a c t i v i t y s t i l l r e m a i n s unknown ( 1 ) . A s c e l l s t r e s s r e s i s t a n c e i s u s u a l l y associated with s t r e s s p r o t e i n synthesis (3»4) our aim was t o s t u d y t h e BS e f f e c t o n p r o t e i n s y n t h e s i s a n d u l t r a s t r u c t u r e o f wheat l e a f c e l l s a t n o r m a l t e m p e r a t u r e a n d u n d e r h e a t s h o c k c o n d i t i o n s . We h a v e a l s o s t u d i e d t h e i n f l u e n c e o f BSs o n m e s o p h y l l c e l l u l t r a s t r u c t u r e under saline stress.

The e f f e c t o f 2 2 S , 2 3 S - h o m o b r a s s i n o l i d e (HBR) on p r o t e i n s y n t h e s i s was s t u d i e d i n t h e f i r s t l e a v e s o f 1 4 - d a y - o l d wheat p l a n t s ov.Saratovskaja 29. The l e a f s e g m e n t s were p r e i n c u b a t e d i n w a t e r o r HBR s o l u t i o n f o r 18 h a t 23 C a n d t h e n i n c u b a t e d f o r 2.5 h a t v a r i o u s t e m p e r a t u r e s ( F i g u r e 1 ). D u r i n g t h e f i n a l 2 h, t h e s e g m e n t s w e r e incubated i n the presence of S-methionine. Uptake of l a b e l e d m e t h i o n i n e by l e a f segments and i t s i n c o r p o r a t i o n i n t o p r o t e i n was analyzed. The percentage of label i n c o r p o r a t e d i n t o p r o t e i n from i t s t o t a l a c c u m u l a t i o n i n l e a f s e g m e n t s was c a l c u l a t e d . T h i s c a l c u l a t i o n a l l o w e d u s t o compare p r o t e i n s y n t h e s i s a c t i v i t i e s i n l e a f segments u n d e r v a r i o u s c o n d i t i o n s (5 ) • ^s Figure shows, HBR a t v e r y l o w c o n c e n t r a t i o n s (10 ^1 a n d 10 M) p r o m o t e d p r o t e i n s y n t h e s i s i n l e a v e s under normal and h i g h temperatures and d e c r e a s e d p r o t e i n synthesis, s e n s i t i v i t y to heat shock. I n the presence o f HBR ( 1 0 ~ M) t h e r a t e o f p r o t e i n s y n t h e s i s a t 43 C was s i m i l a r t o t h a t o b t a i n e d a t 23°C. However, i n u n t r e a t e d l e a v e s p r o t e i n s y n t h e s i s d e c r e a s e d 2.5 f o l d a t 40 C a s compared t o c o n t r o l s a m p l e s ( F i g u r e 1 C ) . HBR a l s o i n d u c e d a l s o some c h a n g e s i n S-methionine uptake by l e a f segments, thus indicating that HBR a f f e c t e d c e l l membranes, e s p e c i a l l y a t 35 C a n d 45 C. A n o t h e r B R ^ 2 4 - e p i b r a s s i n o l i d e ( B B ) , e x e r t e d t h e same e f f e c t on S-methionine a c c u m u l a t i o n i n l e a f segments and i t s i n c o r p o r a t i o n i n t o p r o t e i n (data not p r e s e n t e d ) . While 10~TI EB a t 23 C was much l e s s e f f e c t i v e i n the a c t i v a t i o n of p r o t e i n s y n t h e s i s than HBR. although a t 10" M it was as active as HBR. They both exerted s i m i l a r e f f e c t s on the s e t of polypeptides s y n t h e s i z e d a t 23 C. One-dimensional electrophoresis revealed the a c t i v a t i o n of the s y n t h e s i s of 11 polypeptide groups namely 144. 98. 94. 85. 65. 43. 36-30. 20. 16. 15. 14 kDa. In c o n t r o l leaves p l a c e d under heat shock c o n d i t i o n s (2.5 h, 40 C) the p r o d u c t i o n of the m a j o r i t y of p r o t e i n s diminished and the s y n t h e s i s of heat shook p r o t e i n s (HSPs) was r e v e a l e d . The data concerning HSPs i n r o o t s and c o l e o p t i l e s of 3-day-old wheat s e e d l i n g s (7) and i n a e r i a l p a r t s of 6-day-old e t i o l a t e d wheat s e e d l i n g s (8) were p u b l i s h e d . They I n d i c a t e d the n o n - i d e n t i c a l HSP p a t t e r n s i n v a r i o u s organs (7) o r wheat v a r i e t i e s (8). Moreover the d u r a t i o n of heat shook a l s o a f f e c t e d the HSP p a t t e r n (7). Our experiments d i f f e r e d from (7,8) i n the d u r a t i o n o f heat shook and were performed w i t h the green leaves of 14-day-old wheat s e e d l i n g s (ov.Saratovskaja 29). Thus the complete i d e n t i t y of HSP p a t t e r n s i n our and (7.8) experiments was h a r d l y expected but a c e r t a i n s i m i l a r i t y was r e v e a l e d . Under heat shook c o n d i t i o n s (40 C2gHBR exerted a maximal e f f e c t on p r o t e i n s y n t h e s i s a t 10~ M (Figure 2,8). At 40 C, BSs induced the s y n t h e s i s of the same p o l y p e p t i d e s as those observed at normal temperature (144. 98, 94, 85. 65. 43. 36-30. 20. 16. 15. 14 kDa). Among these polypeptides. 65 kDa protein s y n t h e s i s dominated. I t w i l l be i n t e r e s t i n g to check whether t h i s p r o t e i n corresponds to l a t e HSPs of 63-64 kDa r e v e a l e d i n wheat s e e d l i n g s (7). Molecular weights of some BS-induced p r o t e i n s were i d e n t i c a l to HSPs thus i n d i c a t i n g the p o s s i b i l i t y of BSs p a r t i c i p a t i o n i n the r e g u l a t i o n of HSP s y n t h e s i s . At the same time, BSs s l i g h t l y diminished the s y n t h e s i s of the t y p i c a l HSPs. I t is important that pre treatment of leaves w i t h BSs preserved (at 40 C) the s y n t h e s i s of many polypeptides t y p i c a l f o r normal temperature. T h e p r o d u c t i o n of these p o l y p e p t i d e s was i n h i b i t e d at 40 C i n the c o n t r o l l e a v e s . BSs-induced changes in the sets of proteins s y n t h e s i z e d i n l e a v e s were analysed i n more d e t a i l by two-dimensional SDS-PAAGE (9). Two-dimensional SDS-FAAGE fluorographs of polypeptides s y n t h e s i z e d i n HBR-treated leaves at 23°C and 40°C are shown i n F i g u r e 3 and F i g u r e 4, r e s p e c t i v e l y . At 23°C the production of 180 polypeptides i n c o n t r o l leaves (Figure 3 A) and of 225 polypeptides i n HBR-treated ones (Figure 3 B) were observed. HBR had s t r o n g l y a c t i v a t e d l a b e l i n c o r p o r a t i o n i n t o the 46-65 kDa p r o t e i n f a m i l y s u b d i v i d e d to more than 20 isoforms. HBR a l s o a c t i v a t e d the s y n t h e s i s of some polypeptides l o c a t e d near the RUBISC0 l a r g e subunit (Figure 3 B, L S ) . The frame 1 shows the polypeptides whose s y n t h e s i s was predominantly a c t i v a t e d by HBR. The Q

protein fluorographs, after two-dimensional gel electrophoresis, r e v e a l e d some new spots after HBR treatment that corresponded t o p o l y p e p t i d e s 144 and 36-30 kDa, the l a t t e r a r e I n d i c a t e d by frame 2 i n F i g u r e 3 B. HBR a l s o a c t i v a t e d the formation o f polypeptides 85 and 96 kDa which a r e shown by arrows on F i g u r e 3 B. They corresponded t o HSPs (Figure 3 B, F i g u r e 4 B) The p r e i n c u b a t i o n o f leaves i n HBR s o l u t i o n a l s o r e s u l t e d i n p r o t e i n s y n t h e s i s a c t i v a t i o n under heat shock and t o an i n c r e a s e i n the number o f spots i n the corresponding fluorographs. I t i n d i c a t e d that HBR induced ( o r a c t i v a t e d ) s y n t h e s i s o f a g r e a t number o f p o l y p e p t i d e s under heat shook. The p r o d u c t i o n o f 144 polypeptides i n c o n t r o l leaves and o f 237 Aji leaves incubated i n HBR s o l u t i o n were observed a t 40 G (Figure 4 ) ^ HBR treatment s l i g h t l y diminished HSP s y n t h e s i s a t 40 C and simultaneously Induced s y n t h e s i s of a number o f p o l y p e p t i d e s . Induced by BSs a t normal temperature. Among these p r o t e i n s , p o l y p e p t i d e s o f the 65-46 kDa f a m i l y dominated (Figure 4 B) a t 40 C. A l s o , HBR s t r o n g l y a c t i v a t e d the s y n t h e s i s o f polypeptides l o c a t e d on the fluorographs near the l a r g e subunlt o f RUBISCO. Synthesis of these p o l y p e p t i d e s was i n h i b i t e d a t 40 C i n u n t r e a t e d leaves (Figure 4 A ) . These data i n d i c a t e d that BSs had a protective effect on the formation o f polypeptides typical for normal temperature conditions. Two-dimensional SDS-PAAGE has r e v e a l e d a t l e a s t two p r o t e i n groups, s y n t h e s i s of which was induced o r a c t i v a t e d by both heat shock and HBR. These polypeptides were 98 and 85 kDa (arrows i n F i g u r e 4 B ) . The HBR e f f e c t s on p r o t e i n s y n t h e s i s a t normal and s t r e s s temperatures observed i n wheat leaves o f ov.Saratovskaja 29 were r e v e a l e d a l s o i n our experiments w i t h cv.Opal (data not presented). EB exerted s i m i l a r e f f e c t s on p r o t e i n s y n t h e s i s In wheat leaves as HBR (Figure 5 ) . EB was l e s s a c t i v e i n the stimulation of polypeptide s y n t h e s i s i n wheat leaf segments a t 23°C than HBR and the number o f isoforms induced (or a c t i v a t e d ) by EB was fewer (data a r e not presented). Nevertheless EB was as e f f i c i e n t as HBR i n the a l t e r a t i o n of p r o t e i n s e t s y n t h e s i z e d i n leaves a t 42°C (Figure 5 B ) . Therefore, BSs can induce s i g n i f i c a n t changes i n the activity o f p r o t e i n s y n t h e s i s i n wheat l e a v e s . I n a d d i t i o n , BSs i n c r e a s e d the thermoreslstance o f p r o t e i n s y n t h e s i s under heat shook and p r o t e c t e d the oyntheaia o£ a great number o f polypeptides which were t y p i c a l f o r leaves a t normal temperatures. A c h a r a c t e r i s t i c f e a t u r e of BSs a c t i o n i s the i n d u c t i o n o r a c t i v a t i o n of a number of polypeptides (65-46, 36-30 kDa), s y n t h e s i s . Some of these polypeptides were dominant at normal, and e s p e c i a l l y a t h i g h temperatures. I t i s known that heat shock s t r o n g l y I n h i b i t s p r o t e i n s y n t h e s i s t y p i c a l f o r normal temperatures but i t d i d not a f f e c t BSs-induced

Heat shock i s known t o exert an e f f e c t on p l a n t c e l l u l t r a s t r u c t u r e . The most t y p i c a l change i s represented by the formation o f cytoplasmic heat shock granules (HSG). These s t r u c t u r e s have been observed under heat shock conditions i n many p l a n t c e l l s (10-12). HSGs a r e RNP-particles c o n t a i n i n g temporary (during heat shock) n o n - t r a n s l a t e d mRNAs f o r p r o t e i n s u s u a l l y s y n t h e s i z e d a t normal temperature (13). A l s o , they c o n t a i n two general groups o f p l a n t HSPs: high-molecular p r o t e i n s and mainly low-molecular p r o t e i n s (10). I t i s suggested that HSPs localized i n granules protect mRNAs from heat shook-induced degradation. Because of BS-aotivated s y n t h e s i s o f a number o f polypeptides with molecular weights s i m i l a r t o HSPs, we have i n v e s t i g a t e d the e f f e c t of HBR on HSG formation. F i g u r e 6 A shows the u l t r a s t r u c t u r e o f mesophyll c e l l s i n wheat l e a f segments under heat shock conditions. Electron-dense HSGs aggregated i n small c l u s t e r s and were w e l l d i s t i n g u i s h e d . An average number o f HSGs p e r c l u s t e r was 17 and the average c l u s t e r area was 0.211 rakm- I n l e a f segments pre incubated i n HBR s o l u t i o n (10~ M,18 h) and then subjected t o heat shock (40°C 2.5 h ) , HSG formation was a l s o observed i n the cytoplasm (Figure 6 B). Morphometrio a n a l y s i s has shown that HBR s t i m u l a t e d the HSG formation. The average number o f granules i n c l u s t e r s i n c r e a s e d up to 23 and the average c l u s t e r s i z e extended up t o 0.287 mkm . Therefore HBR s t i m u l a t e d both processes by 40 p e r cent. These r e s u l t s are consonant w i t h the HBR e f f e c t on p r o t e i n s y n t h e s i s under heat shock c o n d i t i o n s . Protective ture

The EB e f f e c t s on c e l l u l t r a s t r u c t u r e were s t u d i e d with the f i r s t l e a f o f 10-day-old b a r l e y s e e d l i n g s (cv.Vingr) Leaf segments were pre incubated i n EB s o l u t i o n (10 «l), o r water, f o r 2 h and were then incubated i n 0.5 M NaCl s o l u t i o n during 24 h i n the presence o r absence o f EB. EB had l i t t l e o r no e f f e c t on the l e a f c e l l u l t r a s t r u c t u r e under normal c o n d i t i o n s (Figures 7 C, 8 C) and s i g n i f i c a n t l y reduced damage Induced by s a l i n e s t r e s s (Figures 7 D, 8 D). Hence EB exerted a p r o t e c t i v e e f f e c t on b a r l e y l e a f c e l l u l t r a s t r u c t u r e under s a l i n e s t r e s s conditions.

BSs p r o t e c t e d c e r e a l l e a f c e l l s from heat shock o r s a l i n e stress. Leaf pre treatment with BSs decreased cell u l t r a s t r u c t u r e degradation from heat shock and h i g h s a l t c o n d i t i o n s . BSs Increased HSG formation which i s supposed to p r o t e c t preformed mRNA i n p l a n t c e l l s during heating. BSs enhanced heat shook resistance of the leaf p r o t e i n - s y n t h e s i z i n g system. The e f f e c t of BSs on RNA and p r o t e i n s y n t h e s i s was shown e a r l i e r (14). We observed p r o t e i n s y n t h e s i s a c t i v a t i o n i n wheat leaves by BSs i n normal and „ under s t r e s s c o n d i t i o n s . Two-dimensional SDS-PAAGE of S-methionine l a b e l e d p r o t e i n s demonstrated BS-induced changes i n the set of polypeptides synthesized i n leaves and i n the r a t e of t h e i r s y n t h e s i s . BSs Induced (or strongly a c t i v a t e d ) s y n t h e s i s of a number of polypeptides. Synthesis of some of them was dominant in leaves under normal and heat shook c o n d i t i o n s . These r e s u l t s permit us to conclude that BSs induced the expression of a number of genes i n wheat l e a v e s . At what l e v e l t h i s r e g u l a t i o n occurs i s s t i l l unknown. The changes i n gene expression were demonstrated i n our experiments a f t e r prolonged p e r i o d of BS a c t i o n (18 h). It i s necessary to find out when these a l t e r a t i o n s occur i n c e l l s and whether they r e l a t e to the previously described changes of membrane s t r u c t u r e / f u n c t i o n (15,16). BS e f f e c t on membranes was a l s o demonstrated i n our experiments by S- methionine uptake by l e a f segments. A l s o , BSs s i g n i f i c a n t l y changed the set of p r o t e i n isoforms, as a p o s s i b l e r e s u l t of their effect on gene expression and (or) post trans l a t i o n a l p r o t e i n m o d i f i c a t i o n . It i s Important that BS-induced changes i n the synthes i z e d polypeptide set were observed not o n l y at normal temperature but a l s o under heat shook c o n d i t i o n s . I t i s known that i n the c e l l s of a l l organisms, from b a c t e r i a to man, the s y n t h e s i s of p r o t e i n s t y p i c a l f o r normal temperature i s I n h i b i t e d by h i g h temperature and synthesis of HSPs i s Induced (3).Some evidence confirm the probable p r o t e c t i v e r o l e of HSPs under s t r e s s c o n d i t i o n s (3,12). The synthesis of BS-induced polypeptides under heat shock g i v e s support to the assumption that BS-induced proteins may a l s o f u l f i l a protective role. 3&
