A new c h r o m a t o g r a p h i c m e t h o d of protein separation based on s o l u t e - m a t r i x interac­ tions is o f f e r e d . A f a m i l y of s u p p o r t s w i t h m i n o r structural variations but significantly different c h r o m a t o g r a p h i c properties al­ lows for selective s e p a r a t i o n . A kit format offers a rapid and simple procedure for selection of the a p p r o p r i a t e gel. Kit I. Alkyl-Agaroses The kit consists of six pre-packed columns containing 1 ml of an alky I agarose of the gen­ eral f o r m u l a A g a r o s e — ( C H J n — H , w h e r e η = 0 , 2 , 4, 6, 8 and 10. Kit II. ω-Amino Alky I Agaroses The kit consists of six pre-packed columns containing 1 ml of an ω-amino alkyl agarose of the general formula Agarose—(CH,) n —NH, where η = 0, 2, 4 , 6 , 8 and 10. Use of these kits facilitates rapid selection of the most a p p r o p r i a t e gel f o r y o u r s y s t e m . Each kit contains fully detailed i n s t r u c t i o n s for use. The c o l u m n s can be re-used c o n ­ tinuously. The individual alkyl-agaroses and ω-amino-agaroses are available in 20 and 50 ml packs.

In 1973 Wilchek d e s c r i b e d the preparation and use of poly-L-lysine agarose and polyDL-alanine poly-L-lysine agarose deriva­ tives. These gels offer the advantages of i n t r o d u c i n g a m u l t i - p o i n t attached hydro­ philic spacer arm. Thus, spacer-ligand l e a c h i n g is r e d u c e d and n o n - s p e c i f i c i n ­ t e r a c t i o n s w i t h the gel m i n i m i z e d . 1. POLY-L-LYSINE-AGAROSE (PL-AGAROSE) Has free a m i n o g r o u p s for c o u p l i n g to ligands c o n t a i n i n g carboxylic g r o u p s . 2. SUCCINYL-PL-AGAROSE (SPL-AGAROSE) Has free carboxyl g r o u p s to enable c o u ­ pling to ligands c o n t a i n i n g free a m i n o groups.

The preparation of hydrazide agarose by a o n e s t e p p r o c e d u r e w a s d e s c r i b e d by L a m e d , Levin and Wilchek in 1973. This gel offers an a m i n e - c o n t a i n i n g spacer s u p p o r t that is free of charges at neutral pH. Al­ t h o u g h p a r t i c u l a r l y s u i t e d to n u c l e o t i d e i m m o b i l i z a t i o n , it can be further derivatized to increase its versatility. The i m p o r t a n c e of u s i n g c h a r g e - f r e e s p a c e r gels in a f f i n i t y c h r o m a t o g r a p h y has r e c e n t l y b e e n d i s ­ cussed by Jost et al. w h o showed that pro­ teins w h i c h b i n d to butyl-agarose d o not bind to butyl hydrazide agarose.

3. POLY-DL-ALANINE POLY-L-LYSINE AGAROSE (PAL-AGAROSE) Multivalent, hydrophilic spacer gel with amino groups far removed from the matrix. 4. SUCCINYL-POLY-DL-ALAIMINEPOLY-L-LYSINE AGAROSE (SPAL-AGAROSE) Carboxyl derivative for coupling to ligands containing free amino groups.

ADIPIC-HYDRAZIDE-AGAROSE Has hydrazide groups on a C e spacer for c o u p l i n g of ligands c o n t a i n i n g free car­ b o x y l i c g r o u p s o r i n s o l u b i l i z a t i o n of periodate oxidized nucleotides.

2. SUCCINYLADIPIC HYDRAZIDE AGAROSE Succinyl derivative with carboxylic g r o u p s for c o u p l i n g to ligands c o n t a i n ­ ing free amino g r o u p s .
