REYNOLDS National Center for Toxicological Research, Jefferson, AR 72074 A multiwavelength, dual beam optical system with 256 detectors operating in a free spectral range of 200 to 800 nm has been designed and implemented for use with a liquid chromatograph. Also, to facilitate the use of this detector an extensive computer software system has been written which allows the user almost unlimited freedom in the way that the data taken over the bandwidth of the detector may be plotted or displayed for visual examination. The linear photodiode arrays used as detectors have been shown to have a significant response in the ultraviolet region of the spectrum. Automation and computerization of laboratory equipment have had a profound effect on the manner in which three important areas of chemistry have been developed and used: 1. The addition of a computer to multiple gas-chromatograph installations permitted rapid data analyses and report generation, 2. The application of Fourier transform methods to nuclear magnetic resonance experiments improved the signal-to-noise ratio and permitted extraction of data heretofore inaccessible. 3. The use of a computer with high-speed high-resolution mass spectrometers allowed vast amounts of data to be collected rapidly and manipulated easily. In all of these cases, the instrument was designed long before the computer was attached. Increasingly, instruments will be designed from the beginning with automation and computerization in mind. The form of the instrument will be determined by the needs of the chemist for proper data, correlated to the needs of the computer. The purpose of this article is to describe an automated unit for liquid chromatography (LC) which utilizes state-of-the-art digital electronics in the instrument and in the associated computer equipment. Many v a r i e t i e s o f r a p i d s c a n n i n g s p e c t r o m e t e r s y s t e m s are available. The f i r s t g e n e r a t i o n u n i t s r e l i e d on moving mechani c a l p a r t s to scan the spectrum. These are t y p i f i e d by the r o t a t i n g m i r r o r or v i b r a t i n g galvanometer types commercially available. The second g e n e r a t i o n i n s t r u m e n t u t i l i z e d V i d i c o n t u b e s o f some s o r t . These e l i m i n a t e d t h e c o n s t r u c t i o n and maintenance problems a s s o c i a t e d w i t h mechanical spectrometers, but t h e h i g h c o s t o f V i d i c o n s y s t e m s , a n d t h e i r t e n d e n c y t o show memory a n d b l o o m i n g makes them i n t e r i m s o l u t i o n s o n l y . The l i g h t t h a t s t r i k e s e a c h e l e m e n t of the a r r a y d u r i n g the time p e r i o d b e f o r e the next s t a r t p u l s e i s i n t e g r a t e d as d e s c r i b e d p r e v i o u s l y . The i n t e g r a t i o n o r exposure time of the a r r a y s i s c o n t r o l l e d b y t h e c l o c k and a s e r i e s o f c o u n t e r s . The minimum i n t e g r a t i o n t i m e a t room t e m p e r a t u r e o f a n a r r a y r u n n i n g w i t h a 20 kHz c l o c k i s 15 m i l l i s e c o n d s ; t h e maximum i n t e g r a t i o n t i m e i s 205 m i l l i seconds. T h i s s i g n a l i s a l s o used i n the f r o n t - e n d processor. Analog Front-End Processor. The sample and r e f e r e n c e v i d e o s i g n a l s a s w e l l a s t h e BOS a n d EOS p u l s e a r e t r a n s m i t t e d v i a c o a x i a l cables to a front-end analog processor ( F i g . 5). The f i r s t s t a g e o f t h e f r o n t - e n d p r o c e s s o r i s a 10 kHz l o w p a s s f i l t e r t o remove n o i s e t h a t i s t h e r e s u l t o f F E T s w i t c h r i n g i n g a s i l l u s t r a t e d i n f i g u r e 6. A f t e r t h e sample and r e f e r e n c e s i g n a l s have been f i l t e r e d the s i g n a l s a r e p l a c e d i n t o v o l t a g e a m p l i f i e r s . The a m p l i f i e r u s e d w i t h t h e r e f e r e n c e s i g n a l may b e u s e d i n e i t h e r a u n i t y or v a r i a b l e g a i n c o n f i g u r a t i o n , c o n t r o l l e d by a two p o s i t i o n s w i t c h t o t h e f r o n t p a n e l . The a m p l i f i c a t i o n i s v a r i a b l e from a g a i n o f 0.5 t o a p p r o x i m a t e l y 2 . 0 . The v a r i a b l e a m p l i f i c a t i o n i s used t o b a l a n c e t h e sample and r e f e r e n c e signals. The o u t p u t f r o m t h e u n i t y g a i n f o l l o w e r s i s i n p u t t o two additional amplifiers. The f i r s t i s a d i f f e r e n t i a l a m p l i f i e r which s u b t r a c t s t h e r e f e r e n c e s i g n a l from t h e sample s i g n a l . The output o f an a r r a y i s a t r a n s m i t t a n c e s i g n a l and i f t h e r e f e r e n c e i s t a k e n t o i n d i c a t e one hundred p e r c e n t t r a n s m i t t a n c e t h e n t h e o u t p u t o f t h e d i f f e r e n t i a l a m p l i f i e r w i l l have a d i r e c t c o r r e spondence w i t h t r a n s m i t t a n c e . The second a m p l i f i e r i n t o w h i c h t h e s a m p l e and r e f e r e n c e s i g n a l s a r e i n p u t i s a l o g - r a t i o amplifier. The o u t p u t o f t h e a m p l i f i e r c a n be c o n s i d e r e d a s absorbance. W i t h i n the f r o n t - e n d processor i s an i n t e g r a t o r that w i l l i n t e g r a t e the output of the l o g - r a t i o - a m p l i f i e r over the p e r i o d between t h e b e g i n n i n g and e n d - o f - s c a n p u l s e s . This i s the e q u i v a l e n t o f t o t a l i o n c u r r e n t i n GC/MS; t h e v a l u e i n L C / U V , V I S i s termed a t o t a l a b s o r p t i o n chromatogram o r T A C . The f r o n t - e n d p r o c e s s o r a l s o p r o v i d e s a v i s u a l warning f l a g when t h e r e f e r e n c e a r r a y s a t u r a t e s . T h i s i s done by c o m p a r i n g the s i g n a l l e v e l r e c e i v e d from the r e f e r e n c e a r r a y w i t h a manually set upper l i m i t . Saturation of the reference array i s n o t a p r o b l e m when t h e d e t e c t o r s y s t e m i s r u n w i t h a r e v e r s e phase o r any s i n g l e s o l v e n t system. T h e p r o b l e m becomes v e r y r e a l when g r a d i e n t e l u t i o n s y s t e m s a r e e m p l o y e d . Optical Design. T h e most common d e s i g n f o r a n u l t r a - v i o l e t s p e c t r o m e t e r i s t h e C z e r n y - T u r n e r m o d i f i c a t i o n o f t h e E b e r t mount for plane grating. T h i s t y p e o f mount was d i s c o u n t e d b e c a u s e t h e d e t e c t o r s b e i n g used r e q u i r e the f a s t e s t o p t i c a l system p o s s i b l e ( f / 3 . 0 o r f a s t e r ) . As p r o v i d e d by O r i e l i t i n c l u d e s a 35 m i l l i m e t e r S u p e r s i l q u a r t z c o n d e n s e r s e t u s e d a s a c o n d e n s e r . A beam s p l i t t e r i s p l a c e d i n t h e l i g h t beam s u c h t h a t i t i s b e t w e e n t h e f o c a l p o i n t o f t h e c o n d e n s e r and t h e c o n d e n s e r . The l i g h t beam i s d i v i d e d i n t o two beams w h i c h w i l l b e u s e d a s a s a m p l e and r e f e r e n c e beam. The s p e c t r o g r a p h s a r e composed o f a c a s t a l u m i n u m h o u s i n g w h i c h c o n t a i n s an e n t r a n c e s l i t , a f r o n t s u r f a c e f o l d i n g m i r r o r , and a c o n c a v e g r a t i n g . The g r a t i n g was d e s i g n e d t o h a v e a f o c a l l e n g t h o f 200 mm and a d i a m e t e r o f 70 mm. T h i s w i l l y i e l d an f-number o f a p p r o x i m a t e l y 3. T h e l i n e s p a c i n g i s 75 l i n e s / m i l l i m e t e r r e s u l t i n g i n a l i n e a r d i s p e r s i o n of 1.2 nanometer p e r one t h o u s a n d t h o f a n i n c h . The u s e o f a c o m p u t e r f o r d a t a a c q u i s i t i o n and c o n t r o l i s mandatory b e c a u s e o f the c l o c k f r e q u e n c y u s e d b y t h e R e t i c o n a r r a y (20 k H z ) and t h e v o l u m e o f d a t a t h a t i t i s capable of o u t p u t t i n g . A P D P - 8 e c o m p u t e r made b y D i g i t a l E q u i p m e n t C o r p o r a t i o n , w i t h 12 K w o r d s o f memory was e m p l o y e d . An E x t e n d e d A r i t h m e t i c ISlement (EAE) i n s t r u c t i o n s e t was installed. Two RK05 m o v i n g h e a d c a r t r i d g e d i s k d r i v e s p r o v i d e mass s t o r a g e c a p a b i l i t i e s of 3.2 m i l l i o n words. The most e c o n o m i c s o l u t i o n to long term storage i s magnetic tape. A 9 track, 800 b i t s p e r i n c h , 45 i n c h e s p e r s e c o n d t a p e d r i v e was u s e d . The ADC c o n v e r t e r h a d a n a n a l o g i n p u t r a n g e o f 0 t o +5 v o l t s a n d a d i g i t a l r a n g e o f 0 t o 1024 (10 b i t s ) . The D / A c o n v e r t e r s u s e d f o r s c o p e d i s p l a y were 10 b i t b i p o l a r c o n v e r t e r s . Two a d d i t i o n a l p i e c e s o f hardware were c o m m e r c i a l l y u n a v a i l a b l e . The f i r s t i t e m was a n e x t e n d e d r a n g e f i x e d p e r i o d c l o c k w i t h a r e s o l u t i o n o f 0.01 seconds and a r a n g e o f 1 6 7 , 7 7 2 . 1 6 s e c o n d s . In u s e , t h i s c l o c k i s s t a r t e d at the b e g i n n i n g o f a chromatographic r u n ; then as each spectrum i s t a k e n f o r s t o r a g e , the time a t w h i c h i t was t a k e n i s r e a d f r o m t h e d a y c l o c k . The s e c o n d h a r d w a r e u n i t was a n i n t e r r u p t , s k i p - c h e c k c o n t r o l u n i t f o r the R e t i c o n a r r a y s . T h i s u n i t a l l o w s the computer t o see the e n d - o f - s c a n and b e g i n n i n g - o f - s c a n p u l s e s g e n e r a t e d by the a r r a y s and a l l o w s the computer to have an o n / o f f c o n t r o l o f the e x t e r n a l c l o c k which i s used to s t a r t the a n a l o g to d i g i t a l converter. The u l t i m a t e g o a l was t o a t t e m p t t o c o r r e l a t e o n c o l o g i c a l events with c e r t a i n " d e s c r i p t o r s " obtained by o b s e r v a t i o n o f t h e U V / V I S r e c o r d . A t t h e o u t s e t , i t was i m p o s s i b l e to d e f i n e the exact r e g i o n s of the s p e c t r a t h a t would be i m p o r t a n t o r how t h e y s h o u l d be d i s p l a y e d b e s t f o r t h e o p e r a tor. I t was e s s e n t i a l t h a t a l l raw d a t a be s t o r e d f i r s t o n d i s k , and s e c o n d on b a c k - u p m a g n e t i c t a p e s o t h a t t h e u s e r c o u l d r e p l a y any g i v e n e x p e r i m e n t i n a v a r i e t y o f i n t e r a c t i v e modes. Theref o r e , s o f t w a r e was d e v e l o p e d w h i c h w o u l d p e r m i t t h e s p e c t r o m e t e r t o be s e t up b y p e r s o n n e l r e l a t i v e l y u n s k i l l e d i n c o m p u t e r o p e r a tion. A f t e r t h e d a t a was c o l l e c t e d i t c o u l d be s c a n n e d b y " c i n e m a t o g r a p h i c " l i k e t e c h n i q u e s , v i e w i n g each successive s p e c t r a i n a f o r w a r d r a p i d s c a n n i n g mode; s t o p p i n g d i s p l a y a t a f r a m e o f i n t e r e s t ; and r e v e r s i n g t h e d i s p l a y . Certain sections o f t h e s p e c t r a c o u l d be i n t e g r a t e d t o p r o v i d e t h e e q u i v a l e n t o f r u n n i n g the experiment w i t h a narrow band p a s s f i l t e r , a l l o w i n g c o m p a r i s o n o f an A b s o r b t i o n Band C h r o m a t o g r a m (ABC) i n c o n t r a s t t o t h e T A C , w h i c h i s an i n t e g r a l o f t h e T o t a l A b s o r b t i o n Chromatogram. T h e ABC c o u l d be c o n s t r u c t e d f r o m a s i n g l e p i x e l , o r t h e e n s e m b l e o f a l l p i x e l s , a t w h i c h p o i n t t h e ABC i s e q u i v a l e n t to the T A C . Any o f t h e s e d i s p l a y s c o u l d be p r e s e n t e d o n a CRT w i t h i n t e r a c t i v e c u r s o r s f o r d e t e r m i n a t i o n o f w a v e l e n g t h o r maxima, and a n y d i s p l a y c o u l d be d i r e c t e d t o a h a r d c o p y d i g i t a l p l o t t e r . T h i s c o n s t i t u t e s a minimum s e t o f s o f t w a r e p r o g r a m s t h a t s h o u l d be p r o v i d e d by any v e n d o r .
