TESTS Sensitivity to Sulfanilamide Dissolve 0.1 g of sample in 100 mL of water to make the sample solution. Then, in two different Nessler tubes, dilute 1.0 and 2.5 mL of sulfanilamide solution (described below) with water to 100 mL. For the blank, omit the sulfanilamide from a third tube, and fill with water to 100 mL. To each tube, add 8 mL of trichloroacetic acid solution and 1 mL of sodium nitrite solution (both described below). After 3 min, add 1 mL of ammonium sulfamate solution (described below), then after 2 additional min, add 1 mL of sample solution. The tubes containing the sulfanilamide should turn pink to red, the higher concentration producing the deeper color.
For the Determination of Sensitivity to Sulfanilamide
S u l f a n i l a m i d e S o l u t i o n . Dissolve 0.02 g of sulfanilamide in 100 mL of water. Tr i c h l o r o a c e t i c A c i d S o l u t i o n . Dissolve 15 g of trichloroacetic acid in 100 mL of water. S o d i u m N i t r i t e S o l u t i o n . Dissolve 0.1 g of sodium nitrite in 100 mL of water. A m m o n i u m S u l f a m a t e S o l u t i o n . Dissolve 0.5 g of ammonium sulfamate in 100 mL of water.
Solubility A solution of 1 g in 50 mL of water is clear, and dissolution is complete.
