Minimizations of compound 1 include surrounding water molecules for the two major conformers of the hydroxymethyl group experimentally detected (gg and gt). As can be seen in these plots and in Table S1, the geometry of the solute significantly changes when decreasing the number of solvating water molecules. When the bridging water molecule is removed (1_gg_11_wat and 1_gg), the carbohydrate and peptide moieties tend to form an intramolecular hydrogen bond, which is not experimentally observed.
