ABSTRACT: The refolding of human serum albumin after unfolding (denaturation) by both acid and base may be followed by a stop-flow method which depends on distinguishing immediate from time-dependent changes in the ionization of its constituent tyrosines at p H values over 10.8 (Steinhardt, J., and Stocker, N. (1973), Biochenzistry 12, 1789). Maximal unfolding at 25” and 0.10 ionic strength occurs a t p H 4 and “swelling” (which alters the viscosity) at p H
