Response to the Letter to the Editor Regarding Our Article (Dheeman et al., 2014)

o the Editor: We appreciate the opportunity to respond to the letter by Professor Naranmandura and thank him for pointing out the similarities between our study and the 2006 Naranmandura et al.1 article. However, we did not discuss his article because it was an analysis of rat metabolites and liver extracts and included no data on AS3MT. The objective of our study was to define the catalytic cycle of the human enzyme AS3MT, and we discussed other reports conducted with purified AS3MT. In our article, we propose a detailed model for the enzymatic reaction that identifies the role of the four conserved cysteine residues and their participation in the formation of enzyme-bound transient intermediates and disulfide bonds that serve to maintain those intermediates in the trivalent oxidation state.
