Using conventional and whole cell patch techniques, we have current- and voltageclamped the somata in cultures of dissociated embryonic cockroach brain neurons and preparations of isolated thoracic neurons from locusts to examine their responses to GABA and other putative neurotransmitters and neuropeptides. These neurons respond to the micro-application of GABA with a transient membrane conductance increase. The effect is blocked by picrotoxin but unaffected by bicuculline up to 10 M. Using the whole cell patch method, we have changed the [Cl ]i of the cockroach neurons, thereby shifting the CI equilibrium potential to demonstrate that the GABA response results from the activation of a CI current. Application of flunitrazepam or Na-pentobarbital to the locust neurons increases the GABA-evoked current by up to70%.However, application of diagnostic agonists and antagonists indicates that these locust somal GABA receptors differ from both the GABA and GABA receptors of the vertebrates. Octopamine, serotonin and FMRFamide all evoke consistent changes in membrane potential and resistance in varying proportions of the cultured cockroach brain neurons. -4

Usherwood and h i s c o - w o r k e r s d e s c r i b e d a t e c h n i q u e f o r i s o l a t i n g n e u r o n a l somata from the t h o r a c i c ganglia of the locust Schistocerca gregaria and, u s i n g a m e t h o d b a s e d on t h e e a r l i e r work o f L e v i M o n t a l c i n i (1), t h e y were a b l e t o c u l t u r e i s o l a t e d n e u r o n a l s o m a t a from l o c u s t s and c o c k r o a c h e s (2,3). A t a b o u t t h e same t i m e , B e a d l e and h i s c o - w o r k e r s d e v e l o p e d a s i m i l a r p r e p a r a t i o n t o show t h a t n e u r o n s f r o m t h e b r a i n s o f embryonic P e r i p l a n e t a americana can be m a i n t a i n e d i n p r i m a r y c u l t u r e f o r many weeks and u n d e r these c o n d i t i o n s produce m u l t i p l e i n t e r - c o n n e c t i n g n e u r i t e s (£,5). In a d d i t i o n , the c u l t u r e d neurons are f r e e o f g l i a and t h u s w e l l - s u i t e d t o p a t c h - c l a m p i n g . B o t h l a b o r a t o r i e s have found t h a t t h e n e u r o n a l somata i n t h e s e p r e p a r a t i o n s a r e endowed w i t h a c o n s i d e r a b l e r a n g e o f r e c e p t o r t y p e s . I n t h i s p a p e r , we w i l l d e s c r i b e o u r r e c e n t o b s e r v a t i o n s on t h e r e s p o n s e s o f t h e c u l t u r e d embryonic c o c k r o a c h b r a i n neurons and f r e s h l y d i s s o c i a t e d l o c u s t t h o r a c i c n e u r o n s t o n e u r o a c t i v e a m i n e s and n e u r o p e p t i d e s , and p a r t i c u l a r l y t h e r e s p o n s e t o y - a m i n o b u t y r i c a c i d (GABA), i t s i o n i c b a s i s and i t s m o d u l a t i o n .

Methods To p r e p a r e t h e l o c u s t n e u r o n a l somata, t h e d e s h e a t h e d t h o r a c i c ganglia of S c h i s t o c e r c a g r e g a r i a or Locusta m i g r a t o r i a were d i s s o c i a t e d m e c h a n i c a l l y , w i t h o u t enzyme treatment, by repeated passage through the t i p o f a P a s t e u r p i p e t t e (2). T i s s u e d i s s e c t i o n , d i s s o c i a t i o n a n d i n t r a c e l l u l a r r e c o r d i n g were c a r r i e d o u t i n a s a l i n e w i t h t h e f o l l o w i n g c o m p o s i t i o n : N a C l , 180 mM; MgCl2, 15 mM; C a C l , 2 mM; KC1, 10 mM; HEPES, 10 mM; pH 6.8. The d i s s o c i a t e d c e l l s were p l a t e d i n a few d r o p s o f s a l i n e o n t o 50 mm " F a l c o n t i g h t - s e a l " p e t r i d i s h e s and l e f t f o r 10 m i n - 4 h r t o a d h e r e t o t h e p l a s t i c s u r f a c e o f t h e d i s h . The r e s u l t i n g m o n o l a y e r o f n e u r o n a l s o m a t a was v i a b l e f o r r e c o r d i n g f o r up t o 12 h r . N e u r o n s f r o m y o u n g e r a n i m a l s were more s t a b l e d u r i n g i n t r a c e l l u l a r r e c o r d i n g . The c e l l s r a n g e d b e t w e e n 10 and 100 yum i n d i a m e t e r and o f t e n b o r e a x o n stumps w i t h a p p a r e n t r e g i o n s o f r e g e n e r a t e d membrane a t t h e i r t i p s . O n l y t h e l a r g e r n e u r o n s (30-100/im) were s e l e c t e d f o r i m p a l e m e n t . C o n v e n t i o n a l r e c o r d i n g and s i n g l e e l e c t r o d e v o l t a g e c l a m p i n g methods were u s e d . The p r e p a r a t i o n o f t h e e m b r y o n i c c o c k r o a c h b r a i n neuronal c u l t u r e s i s a complicated procedure which has b e e n d e s c r i b e d i n d e t a i l b y B e a d l e and L e e s ( £ ) . I n b r i e f , t h e b r a i n s were d i s s e c t e d f r o m 23-26 d a y o l d embryos o f P e r i p l a n e t a a m e r i c a n a and b r o k e n up b y p a s s a g e t h r o u g h P a s t e u r p i p e t t e s o f d e c r e a s i n g d i a m e t e r . The c e l l s w e r e grown a t 29°C i n 50 mm " F a l c o n t i g h t s e a l petri d i s h e s u s i n g a m o d i f i c a t i o n o f t h e h a n g i n g d r o p method. Discussion

C u l t u r e d cockroach neurons. S e v e r a l b i o g e n i c amines, n e u r o p e p t i d e s and o t h e r p u t a t i v e n e u r o t r a n s m i t t e r s were a p p l i e d by p r e s s u r e e j e c t i o n from m i c r o p i p e t t e s onto t h e s u r f a c e o f i n d i v i d u a l n e u r o n a l somata under c u r r e n t c l a m p . The r e s u l t s a r e i l l u s t r a t e d i n F i g . 1 and h a v e b e e n r e p o r t e d b r i e f l y e l s e w h e r e (2). A l l o f t h e c e l l s t e s t e d (N>50) r e s p o n d e d t o a c e t y l c h o l i n e w i t h a v o l t a g e d e p e n d e n t d e p o l a r i z a t i o n and a d e c r e a s e i n membrane r e s i s t a n c e , a s d e s c r i b e d i n d e t a i l b y L e e s Ê£ al(S)· I n a d d i t i o n , 60% o f t h e c e l l s were s e n s i t i v e t o GABA, r e s p o n d i n g w i t h a d e c r e a s e i n membrane r e s i s t a n c e and a c o n c u r r e n t h y p e r p o l a r i z a t i o n , e x c e p t i n somata w i t h v e r y n e g a t i v e r e s t i n g p o t e n t i a l s (more n e g a t i v e t h a n -55 t o -65 mV) w h e r e t h e r e s p o n s e was a d e p o l a r i z a t i o n ( F i g . l a ) . The GABA r e s p o n s e w i l l b e c o n s i d e r e d i n g r e a t e r d e t a i l below. Octopamine evoked h y p e r p o l a r i z a t i o n s a c c o m p a n i e d b y membrane r e s i s t a n c e i n c r e a s e s i n 2 o f 9 neurons t e s t e d . I t slowed the r a t e of s p o n t a n e o u s s p i k i n g where t h i s o c c u r r e d ( F i g . l b ) and t h e e v o k e d h y p e r p o l a r i z a t i o n was l a r g e r a t more n e g a t i v e membrane p o t e n t i a l s ( F i g . l c ) . S e r o t o n i n ( 5 - h y d r o x y tryptamine) evoked d e p o l a r i z a t i o n s accompanied by d e c r e a s e d membrane r e s i s t a n c e ( F i g . I d ) , w i t h t h e e v o k e d p o t e n t i a l s i n c r e a s i n g i n magnitude w i t h h y p e r p o l a r i z a t i o n o f t h e membrane p o t e n t i a l . R e p e a t e d p u l s e s o f s e r o t o n i n r e s u l t e d i n a response o f d e c r e a s i n g magnitude ( F i g . l e ) s u g g e s t i n g r e c e p t o r d e s e n s i t i z a t i o n . Glutamate evoked r e s p o n s e s i n up t o 30% o f t h e n e u r o n s i n some c u l t u r e s . The m a j o r i t y o f t h e r e s p o n s e s were d e p o l a r i z a t i o n s a c c o m p a n i e d b y d e c r e a s e s i n membrane r e s i s t a n c e b u t some c e l l s were h y p e r p o l a r i z e d (Horseman eJt a l . , unpublished d a t a ) . U s i n g an i n t r a - e l e c t r o d e [CI"] o f 114 mM and [C1"] o f 221 mM, t h e E R o f t h e GABA-evoked c u r r e n t was s h i f t e d t o -16 mV ( F i g . 2) w h i c h i s e q u a l t o t h e v a l u e p r e d i c t e d f o r Eci b y t h e N e r n s t e q u a t i o n . From t h e s e e x p e r i m e n t s , we c o n c l u d e t h a t t h e GABA r e s p o n s e o f t h e c u l t u r e d embryonic cockroach b r a i n neurons i s mediated by C I " i o n s , a s i n t h e v e r t e b r a t e GABA^ s y s t e m , and t h a t r a p i d e q u i l i b r a t i o n o c c u r s between the i n t r a - e l e c t r o d e and i n t r a c e l l u l a r [CI"] when t h e w h o l e c e l l p a t c h c l a m p method i s employed.
