ProSwift Monolith Columns Deliver Fast Protein Analysis Monolith columns containing an uninterrupted network of interconnected channels are proving to be an important new technology for protein analysis because the monolith structure supports fast analyte mass transfer. Small molecules are quickly separated using conventional porous silica and polymer bead packing in HPLC columns, but proteins and other large biomolecules diffuse slowly, resulting in broad peaks. Convective flow of the monolith is superior to the slow mass transfer by diffusion associated with traditional columns packed with porous beads. ProSwift Monolith columns, from Dionex Corporation, feature a unique monolith morphology that delivers high permeability, low backpressure, and high flow rates,
resulting in high-speed and highresolution HPLC separations of proteins and other large biomolecules. ProSwift columns therefore have higher productivity than columns packed with conventional porous silica and polymer beads. The monoliths provide excellent chemical stability, allowing use of a wide pH range for separation and regeneration. Monolith resin is highly cross-linked for physical stability and long column life.
