The lsomerlc octopamlnes and synephrines were measured In urine by a new assay which combines ion-exchange chromatography, capillary column gas chromatography, and electron capture negative ion chemical ionization mass spectrometry. Deuterium labeled analogues of each compound were added to urlne contalnlng 1 mg of creatlnlne and the acid hydrolysate was subjected twice to cation exchange chromatography. The resultant amlne fraction was derlvatired wlth pentafluoroproplonlc anhydrlde and, under electron capture negative chemical Ionization condltlons, the PFP derivatives gave M- and (M HF)- Ions which were sufflciently abundant to be sukable for selected ion monltorlng. The ilmlt of detection was approximately 100 pg mg-’ creatinine. I n 10 normal adults, the concentrations of 0 - , m-, and p-octopamlne and 0-, m- and p-synephrlne were, respectlvely, 0.6 (*0.2), 2.1 (f0.8), 25 (flO), ND (
