Rapid Extraction and Detection of Aflatoxin M, in Cow’s Milk by High-Performance Liquid Chromatography and Radioimmunoassay Geng-Sun Qian,’ Parvin Yasei,2 and George C. Yang* Food and Drug Administration, Division of Chemistry and Physics, Washington, D.C. 20204
A procedure was developed for determining aflatoxln M, in cow’s mllk, using a simple and rapld solld-phase extractlon with a silica gel C,, dlsposable column In conjunction wlth normal-phase hlgh-performance liquid chromatography (HPLC) wlth a silica gel packed flow cell fluorescence detector and radiolmrnunoassay (RIA). Thls procedure allows the dlrect loading of milk on the dlsposable column at the dairy farm or processing facility. M,, which is adsorbed on the column, is recovered by elutlon with 5 % acetone In methylene chloride. The recovery levels of M, In spiked mllk Samples from columns stored at amblent temperature for 1 week did not change. The limit of measurement In splked milk (10-mL samples) was
