ABSTRACT This monograph for Glycine provides, in addition to common physical constants, a general description including typical appearance, applications, change in state (approximate), and aqueous solubility. The monograph also details the following specifications and corresponding tests for verifying that a substance meets ACS Reagent Grade specifications including: Assay, Residue after Ignition, Heavy Metals, Chloride, Sulfate, Ammonium, Substances Darkened by Sulfuric Acid, and Hydrolyzable Substances.

TESTS Assay (Total alkalinity by nonaqueous titration). Weigh accurately 0.25 g, and dissolve in 100 mL of glacial acetic acid. Add 0.5 mL of crystal violet indicator solution, and titrate with 0.1 N perchloric acid in glacial acetic acid volumetric solution to a green end point. Correct for a blank. One milliliter of 0.1 N perchloric acid corresponds to 0.007507 g of H2NCH2COOH. % H 2NCH 2COOH =

{[mL (sample)

mL (blank)] × N HClO4 } × 7.507 Sample wt (g)

Residue after Ignition [Part 2: Gravimetric Methods; Residue after Ignition]. Use 2.0 g.

Heavy Metals [Part 2: Colorimetry and Turbidimetry; Heavy Metals (as Lead); Procedure for Heavy Metals, Method 1]. Use 1.0 g.

Chloride [Part 2: Colorimetry and Turbidimetry; Chloride]. Use 0.2 g.

Sulfate [Part 2: Colorimetry and Turbidimetry; Sulfate; Procedure for Sulfate, Method 1]. Use 1.0 g.

Ammonium (By colorimetry, [Part 2: Colorimetry and Turbidimetry; Ammonium (Test for Ammonia and Amines)]). Use 0.2 g. For the standard, use 0.01 mg of ammonium ion (NH4) treated exactly as the sample.

Substances Darkened by Sulfuric Acid Dissolve 0.5 g in 5 mL of sulfuric acid. Allow the solution to stand for 15 min. The color of the sample solution should not exceed that of an equal volume of the acid.

Hydrolyzable Substances Boil 10 mL of a 1 in 10 solution of glycine for 1 min, and set aside for 2 h. The solution, when compared to an equal volume of an untreated sample solution, should appear as clear and as mobile.
